ADVANCE
11.140 patients, 214 sites, finalised 2008 Together with the AACT partners, The Action in Diabetes and Vascular Disease: Preterax and Diamicron Modified Release (MR) Controlled Evaluation (ADVANCE) trial was performed. This phase III clinical trial was conducted in 214 collaborating centers in more than 20 countries worldwide.
The ADVANCE trial was designed to provide reliable evidence about benefits and risks conferred by routine blood pressure lowering therapy and intensive glucose control therapy in high-risk type 2 diabetic patients, irrespective of initial blood pressure or glucose levels. The primary endpoints were composites of major macrovascular and microvascular events, analysed jointly and separately by intention to treat.
After 6 weeks on open label perindopril-indapamide combination, eligible individuals were randomised to continued perindopril-indapamide or matching placebo, and to an intensive gliclazide MR-based glucose control regimen or usual guidelines-based therapy.
The Julius Center coordinated the European part of the investigation. Commenced in June 2001, successful worldwide recruitment was completed in March 2003, with the inclusion of 11.140 high-risk individuals with type 2 diabetes.
This trial is registered on ClinicalTrials.gov, number NCT00145925.
